Cannabinoids Reduces Migraine-Like Symptoms in Preclinical Study (pub. Jan 2026)

Migraines are often linked to increased activity of calcitonin gene-related peptide (CGRP), a signaling molecule known to trigger pain sensitivity, light sensitivity, and other migraine symptoms. In this preclinical study, researchers examined whether a combination of cannabidiol (CBD) and tetrahydrocannabinol (THC) could reduce migraine-like symptoms caused by central CGRP administration in mice. Scientists used established behavioral models that mimic migraine features such as light aversion (photophobia), reduced activity, and spontaneous pain responses. (PubMed)

The results showed that pretreatment with a high-CBD, low-THC formulation (100:1 ratio) significantly improved several migraine-related behaviors. Mice receiving the cannabinoid combination spent more time in lighted environments, indicating reduced light sensitivity, and showed improvements in movement and exploratory behavior that were previously suppressed by CGRP exposure. The treatment also partially reduced spontaneous pain responses measured through facial expression and behavioral testing. Importantly, the study found no major impairments in anxiety, cognition, or motor performance associated with the cannabinoid treatment. (PubMed)

Overall, the findings suggest that cannabinoids may influence migraine pathways by counteracting CGRP-driven mechanisms involved in pain and sensory sensitivity. While these results come from animal models rather than human clinical trials, the research provides early evidence supporting further investigation of cannabinoid-based therapies as potential supportive approaches for migraine management.
